Head Coach Richard Hill makes a total of six changes to the side that started the 22-7 victory over Scarlets on Saturday afternoon with Schofield returning to partner James Percival in the second row.
The other change in the pack comes in the back row where flanker Matt Kvesic is given his first start of the pre-season campaign.
Scrum half Shaun Perry and fly half Andy Goode are reunited at half back for the clash against the RFU Championship outfit while Chris Pennell is recalled at full back with Errie Claassens moving to the wing.
Fiji ace Josh Matavesi, fresh from his two-try heroics against Scarlets at the weekend, is paired with exciting England Under-18 centre prospect Max Stelling in midfield as Hill continues to look at his back line options.
Prop George Porter is named among the replacements while Hill also gives the next generation a potential chance to impress from the bench with back row trio Andrew Boyce, Nile Dacres and Ed Taylor, centre Jack Wardle, wing Joe Nellany and fly half Tom Carlisle all selected. Winger Ed Tellwright makes a guest appearance for the club from the bench.
"We want to keep building momentum ahead of the new Aviva Premiership season and so have picked a strong side as we look to get better as a unit and continue to improve our performances," said Hill.
"Rotherham will provide a stern test on home soil, as we know from our season in the RFU Championship, but I am looking for the Warriors to pick up where we left off against Scarlets and raise the bar once more."
